摘 要

毛细管电色谱作为一种新型的色谱分离技术,同时兼具了高效液相色谱的高选择性和毛细管电泳的高分离效率,在分离分析对映体上极具潜力。多糖,如淀粉,因具有空间高度规则的螺旋结构,被广泛用作手性固定相。本实验以淀粉手性填料为手性固定相制作毛细管色谱柱,探讨电色谱对氧氟沙星对映体手性拆分的条件,主要包括磷酸盐溶液的pH值,缓冲溶液的组分比例,分离电压等。但在目前实验条件下,氧氟沙星对映体拆分效果不明显。未来将从手性固定相的选择,流动相的选择,加入手性添加剂等方面进行改进。

Abstract

Capillary electrochromatography, As a new chromatographic separation technology, combines the high selectivity of high performance liquid chromato- graphy with the high separation efficiency of capillary electrophoresis, and has great potential for the separation and analysis of enantiomers. Polysaccharides, such as starch, have been widely used as chiral stationary phases because of their spatially regular helix structure. In this experiment, the capillary chiral column was used as the chiral stationary phase to study the conditions for the chiral separation of ofloxacin enantiomers, mainly including the pH of the phosphate- solution and the proportion of the buffer solution, separation voltage, etc. However, under the current experimental conditions, the effect of enantiomer separation of ofloxacin is not obvious. The future will be improved from the selection of chiral stationary phases, the selection of mobile phases, and the addition of chiral additives.
