氨基化多孔硅的制备及应用

摘 要

多孔硅作为一种具有纳米硅原子簇为骨架的量子海绵状微结构的新型的纳米材料。自1956年它首次进入研究人员视野后,因为它具有比面积大、生物相容性好、良好的发光特性等特点,被广泛应用于与光学成像、抗菌、抗癌药物载体、质粒转染等方面。但是在作为药物载体方面,由于多孔硅表面不具有封闭空隙的聚合物,导致其负载的颗粒在生物环境中爆裂释放或提前释放,导致部分区域药物浓度急剧提升,达到对生物体有害的程度,并且药物不能有效的到达理应释放的区域。对于多孔硅表面进行化学修饰和与金属离子配位,可以大大提高多孔硅的药物载荷量。本文通过多孔硅的氨基化和在此基础上与金属离子Mn2 、Ag 、Mg2 配位,提高对抗癌药物阿霉素的药物载荷量。

Preparation and Application of Aminated Porous Silicon

Abstract

Porous silicon is a new type of nanomaterial with a quantum sponge-like microstructure with nano-silicon atom clusters as a skeleton. Since it first entered the field of vision of researchers in 1956, it has been widely used in optical imaging, antibacterial, anticancer drug carriers, plasmid transfection, etc. because of its large specific area, good biocompatibility, and good luminescence characteristics. However, as a drug carrier, the porous silicon surface does not have a polymer with closed voids, which causes its loaded particles to burst or release early in the biological environment, leading to a sharp increase in drug concentration in some areas, which is harmful to the organism. And the drug cannot reach the area where it should be released effectively. Chemical modification of the surface of porous silicon and coordination with metal ions can greatly improve the drug loading capacity.In this paper, through the amination of porous silicon and coordination with the metal ions Mn2 , Ag , and Mg2 on this basis in order to increase drug loading of anticancer drug doxorubicin.
